After a week, the contents of capsules, which contain the following raw materials: Pheniramine Maleaat 35mg Triamcinolone 4mg pseudoephedrine HCl 20mg

After 1 week, this combination results in a kind of gel and the capsule look bad. We have used lactose as a filler (Gelule nr 4) and do not know how to improve that preparation.

first possibility
  1. Choose a larger size of capsules eg. No 2. This will allow you to dilute the medicines with lactose.
  2. determine how much (in ml) lactose is required to fill the number of capsules to be prepared for the chosen size. E.g. x ml
  3. Dilute the mixture Pheniramine + triamcinolone with 3g lactose and measure the volume of the mixture (A). Withdraw this volume from X. Outcome = Y
  4. Dilute pseudo-Ephedrinehcl with lactose ensuring that the volume of this mixture (B) does not exceed the value Y!!
  5. Mix the mixtures A and B and fill the total mixture in the capsules.
Second possibility in order to avoid any interaction between pheniramine and pseudoephedrine, it is of course appropriate to separate the 2 substances completely from each other by wrapping them in two different capsules.
for this we transfer the mixture Pheniramine + triamcinolone into a capsule Nr 3. This capsule has an internal content of 0.27 ml. The total volume wall + internal space is estimated at 0.3 ml. If we use e.g. a capsule 0 for filling the pseudoephedrine HCl, we have a free space of (0.67-0.3) = 0.37 ml. This free space can be used to replenish pseudoephedrine HCl diluted with lactose. In the preparation of this last mixture, consideration should be given to not exceeding the volume of 0.37 ml per capsule.
